Identification of a marine agarolytic Pseudoalteromonas isolate and characterization of its extracellular agarase
Article Abstract:
A new agarolytic bacterium from the southern Chilean coast was characterized and determined to be Pseudoalteromonas antarctica N-1 by phylogenetic studies based on analysis of the 16S rRNA gene sequence. The isolate produced a diffusible agarase which induced agar softening around the colonies. An extracellular agarase was purified and found to be homogeneous. The enzyme was able to hydrolyze the beta-1,4-glycosydic linkages of agar, producing neoagarotetraose and neoagarohexaose as the primary products.

Article Abstract:
Isolation and characterization of a novel haloacid permease from Burkholderia cepacia is described. The structural gene deh4p was found 353 bases downstream of the dehalogenase gene deh4a and these two genes form an operon that mediates the utilization of haloacetate in Burkholderia cepacia MBA4.

Seventeen amino acid residues between position 73-74 and 89-90 determine whether haloacid dehalogenases from Burkholderia cepacia aggregate as a monomer or a dimer. In addition, Phe58, Thr65, Leu78, and Phe92 are also important.
