Identification of the functional subunit of a dimeric transcription activator protein by use of oriented heterodimers
Article Abstract:
Escherichia coli catabolite gene activator protein (CAP) is a dimer with two identical subunits, each having two activating regions. CAP activates transcription at the lac promoter. Oriented heterodimers were used to determine which activating region is needed for transcription activation at the lac promoter. Results show that the promoter-proximal subunit is the only activating region needed for transcriptional activation.

Inhibition of bacterial RNA polymerase by streptolydigin: Stabilization of a straight-bridge-helix active-center conformation
Article Abstract:
Streptolydigin (Sti), the tetramic acid antibiotic, binds to a site adjacent to the RNA polymerase (RNAP) active center and stabilizes an RNAP-active-center conformational state with a straight-bridge helix. The results support the proposals that alternative straight -bridge-helix and bent-bridge-helix RNAP-active-center conformations exist and cycling between the two is required for RNAP function.

Translocation of sigma(sup)70 with RNA polymerase during transcription: fluorescence resonance energy transfer assay for movement relative to DNA
Article Abstract:
Results demonstrate that the sigma(sup)70 in the transcription complexes largely are not released from RNA polymerase upon transition from initiation to elongation during genetic transcription in bacteria. Data indicate that sigma(sup)70 stays associated with and translocates with RNA polymerase.
