In situ PCR for visualization of microscale distribution of specific genes and gene products in prokaryotic communities
Article Abstract:
A new method based on prokaryotic in situ polymerase chain reaction (PI-PCR) provides information about specific genes and gene transcripts in bacterial cells, which can be used to identify individual prokaryotic species. The method involves amplification of nucleic acid sequences inside prokaryotic cells followed by color or fluorescence identification of the PCR product by bright-field or epifluorescence microscopy. The method is tested by identifying nahA-positive cells in marine model or mixed bacterial cultures.

Direct extraction and purification of rRNA for ecological studies
Article Abstract:
The utilization of 16S rRNA probes for RNA extraction from soil, sediment and water samples is discussed. This involved the utilization of a lysozome-hot phenol direct eextraction method. Organic contaminants that interfere with subsequent molecular procedures were also derived with the recovery of either DNA or RNA from natural samples. A modified gel filtration technique was able to remediate the RNA extracts for sufficient molecular procedure quality.

Evidence for indigenous Streptomyces populations in a marine environment determined with a 16S rRNA probe
Article Abstract:
Populations of Streptomyces bacteria appear to inhabit coastal marshes indigenously rather than be transplanted from terrestrial communities. Using a 16S rRNA probe to detect bacteria of the genus Streptomyces, researchers found the bacteria in several samples of sediment from marshes along the Georgia coast. Bacteria in this genus constituted 2% to 5% of the bacterial community in the marsh sediment.
