Insertional mutations in the hydrogenase vhc and frc operons encoding selenium-free hydrogenases in Methanococcus voltae
Article Abstract:
The inability of the mutated vhc and frc operons to form any (NiFe)-enzymes in Methanococcus does not cause any cell death in the laboratory even in the absence of selenium. The selenium-free enzymes are produced when selenium is absent. However, it is possible for small concentrations of (NiFeSe)-hydrogenases to induce the formation of the (NiFe)-enzymes. In laboratory conditions, hydrogen does not affect the growth of the cells, but it does so in the wild, so it is possible for the cells' inability to form the (NiFe)-enzymes to cause their death in the wild.

Formation and regeneration of Methanococcus voltae protoplasts
Article Abstract:
A method for the protoplastic conversion of Methanococcus voltae cellsis discussed. The formation of protoplasts was microscopically monitored as gelelectrophoresis and electron microscopy determined the formation of the protoplasts. Optimized plating conditions and an anaerobic microplating technique evaluated protoplast regeneration. The utility of the method was demonstrated.

Pressure stabilization is not a general property of thermophilic enzymes: the adenylate kinases of Methanococcus voltae, Methanococcus maripaludis, Methanococcus thermolithotrophicus, and Methanococcus jannaschii
Article Abstract:
The stabilization by pressure is not an inherent property of the structures of all thermophilic enzymes isolated from thermophilic marine methanogens. The thermostabilities of adenylate kinases obtained from mesophiles Methanococcus voltae and M. maripaludis, and thermophile M. jannaschii and M. thermolithotrophicus, are not increased by employing a pressure of 50-MPa.
