Intermediates in formation and activity of the RNA polymerase II preinitiation complex: holoenzyme recruitment and a postrecruitment role for the TATA box and TFIIB
Article Abstract:
Activity and assembly of yeast RNA polymerase II (Pol II) preinitiation complexes (PIC) have been investigated. The investigation involved an immobilized promoter assay and extracts made from wild-type cells and cells having conditional mutations in parts of the Pol II mechanism. In one step TFIID and RFII assemble at the promoter independently of the holoenzyme and, in another, holoenzyme is recruited to the promoter. With other findings, an involvement of the TATA box and TFIIB has been shown in one of more steps after recruitment of factors to the promoter.

Extensive post-transcriptional regulation of microRNAs and its implications for cancer
Article Abstract:
A large fraction of microRNAs genes are regulated post-transcriptionally and an analysis of these gene expressions in primary tumors indicates that the widespread downregulation of microRNAs cancer is due to a failure at the Drosha processing step. These data uncover a regulatory step in microRNA function and provide a mechanism for microRNA down-regulation in cancer.

Positive and negative functions of the SAGA complex mediated through interaction of Spt 8 with TBP and the N-terminal domain of TFIIA
Article Abstract:
A surface that is required for rapid formation of preinitiation complexes (PICs) was identified on the N-terminal domain (NTD) of the RNA Pol II general transcription factor TFIIA. A study was conducted and the results suggested a mechanism for the previously observed positive and negative effects of Spt 8 on transcription observed in vivo.
