Linkage analysis of human leukocyte antigen (HLA) markers in familial psoriasis: strong disequilibrium effects provide evidence for a major determinant in the HLA-B/-C region
Article Abstract:
Linkage analysis has been carried out on human leukocyte antigen (HLA) markers in familial psoriasis. Sporadic and familial psoriasis have the same risk alleles and it can be seen that substantial parametric linkage information can be had by accounting for linkage disequilibrium. It seems a major susceptibility gene is near HLA-C. Strong disequilibrium effects have provided evidence for a major determinant in the HLA-B/-C region. Evidence for linkage to HLA markers has been limited although psoriasis is strongly associated with certain human leukocyte antigens.

A study to identify the PSORS1 gene, the psoriasis-susceptibility locus in the major histocompatibility complex (MHC) is presented. The results suggest that HLA-Cw6 is the PSORS1 risk allele that confers susceptibility to early-onset psoriasis.
