Microbial proline 4-hydroxylase screening and gene cloning
Article Abstract:
The partial purification, proline 4-hydroxylase screening, cloning and expression of the proline 4-hydroxylase gene in Escherichia coli were conducted. The proline 4-hydroxylase studied is classified as a 2-oxoglutarate-dependent dioxygenase since it requires 2-oxoglutarate and ferrous ion for a reaction. It catalyzed highly regio- and stereospecific hydroxylation of L-proline. The rigidity of the reaction proves the usefulness of the enzyme as a biocatalyst for the production of trans-4-hydroxy-L-proline from L-proline.

Construction of a reporter vector for the analysis of Bifidobacterium longum promoters

A versatile reporter plasmid based on a Bifidobacterium longum cryptic plasmid and the Escherichia coli gusA gene is constructed to initiate studies on promoter activities in Bifidobacterium langum and to independently confirm transcriptional data generated by microarray experiments. The resulting plasmid, pMDY23 is tested using three B. longum promoters.
