Neurospora: the organism behind the molecular revolution
Article Abstract:
Research on the bread mold Neurospora has contributed much to the development of genetics as a discipline. First described in 1842, its behavior had been the subject of microbiological research before its utility in genetics was demonstrated. Study of Neurospora resulted in the discovery of the mechanisms of fungal reproduction, meiosis and recombination. It was also used in the elucidation of the one-gene, one-enzyme hypothesis. This mold was ideal for research use due to its haploid character, ease of culture and rapid growth. Although widely used in research, its evolutionary biology and population genetics still require further study.

Analysis of junction sequences resulting from integration at nonhomologous loci in Neurospora crassa
Article Abstract:
The DNA sequences at junctions between transforming and chromosomal DNA from two ectopic integration events were analyzed. The absence of homology at the junctions suggests that the integrations involved illegitimate recombination and resulted from the end joining of a truncated, linearized plasmid to a preexisting break in chromosomal DNA. The transforming DNA in one of the events linked sequences from two different chromosomes, thus bridging a translocation.

Chromosome rearrangements recovered following transformation of Neurospora crassa
Article Abstract:
An analysis of molecularly characterized am+ transformants revealed that realignments occur often when multiple ectopic integration events occur, but fail to occur at all or as often when only the resident locus is restored to am+ by a homologous event. Breakpoints in six of 10 translocations, analyzed with southern hybridization or colony blots, were genetically associated with sequences of the changing vector.
