Rad54 and DNa Ligase IV cooperate to maintain mammalian chromatid stability
Article Abstract:
The cooperation of Lig4 and Rad54 to support cellular proliferation, repair of spontaneous DSBs, and prevention of chromosome and single chromatid aberrations was demonstrated. The findings showed a role for nonhomologous end joining (NHEJ) in the repair of double-strand breaks (DSBs) that occur spontaneously during or after DNA replication, and reveal overlapping functions for NHEJ and Rad54-dependent HR in the repair of such DSBs.

Genomic instability and aging-like phenotype in the absence of mammalian SIRT6
Article Abstract:
Silent information regulator (SIRT6)-deficient mice are small and at 2-3 weeks of age develop abnormalities that include profound lymphopenia, loss of subcutaneous fat, lordokyphosis, and severe metabolic defects, eventually dying at about 4 weeks. It is concluded that one function of SIRT6 is to promote normal DNA repair, and that SIRT6 loss leads to abnormalities in mice that overlap with aging-associated degenerative processes.

Histone H2AX: a dosage-dependent suppressor of oncogenic translocations and tumors
Article Abstract:
Results point out that the phosphorylated histone variant H2AX prevents aberrant repair of both programmed and general DNA breakage, suggesting that H2AX functions as a suppressor of genomic instability and tumors in mice. Furthermore, H2AX is localized to a cytogenetic region that undergoes frequent changes in human cancers.
