Randomly amplified polymorphic DNA analysis of starved and viable but nonculturable Vibrio vulnificus cells
Article Abstract:
A new randomly amplified polymorphic DNA (RAPD) polymerase chain reaction method for the detection of starved and viable but nonculturable Vibrio vulnificus cells has been devised. Changes in the RAPD profile can be detected as early as 15 minutes into the starvation period when the Vibrio cells are subjected to starvation in artificial seawater. There was also a uniform loss of RAPD amplification products. Cells that have been starved for up to one year were detected again when nutrients were added to the starvation microcosm.

A rapid method for extraction and purification of DNA from dental plaque
Article Abstract:
DNA isolated from dental plaque by a new method is suitable for analysis by polymerase chain reaction (PCR). The purified DNA is free from PCR inhibitors. The method involves isolating DNA from garm-positive and gram-negative bacteria in uncultured dental plaque and yields highly purified DNA. The protocol comprises cell lysis by freeze-thawing and by treating with cell wall degrading enzymes to liberate the DNA, adsorption of DNA onto diatomaceous earth, and washing by guanidine isothiocyanate, acetone and ethanol.
