Repair of oxidative DNA damage in Gram-positive bacteria: the Lactococcus lactis Fpg protein
Article Abstract:
Molecular cloning and sequencing of the formamidopyrimidine DNA glycosylase gene (fpg-L) of the gram-positive microaerophilic bacterium Lactococcus lactis subsp. cremoris ML3 reveal the significant similarity between the amino acid sequence of Fpg-L protein and that of Escherichia coli Fpg protein (Fpg-E). This similarity suggests a common ancestor for the fpg-L and the fpg-E. These proteins protect bacterial DNA against oxidative free radicals. Active Fpg-L has one zinc per molecule and the zinc finger motif in Fpg-L is localized at the carboxy-terminal end.

Use of degenerate primers for polymerase chain reaction cloning and sequencing of the Lactococcus lactis subsp. lactis recA gene
Article Abstract:
Degenerate primers of DNA were designed using conserved stretches of amino acids of known RecA proteins. The primers included an internal fragment of the Lactoccocus lactis subsp. lactis recA gene. Amplification was done using the polymerase chain reaction, and the entire lactococcal recA gene was recovered and sequenced. The lactococcal gene had homology to recA genes from Bacillus subtilis and Escherichia coli. An open reading frame upstream of the gene showed strong homology to formamidopyrimidine-DNA-glycosylase, a DNA repair enzyme.

Divergence of genomic sequences between Lactococcus lactis subsp. lactis andLactococcus lactis subsp. cremoris
Article Abstract:
New means of examining the relation between the Lactoccus lactis subspecies cremoris and lactis are presented. Southern hybridization analysis using cloned chromosomal genes as probes was used to assess the bacteria. It was found that two distinctgroups are formed by the two subspecies while a 20 to 30% sequence divergence is estimated between the two. This DNA homology is the basis for the call for anew classification between the groups.
