Seasonal and spatial variability of bacterial and archaeal assemblages in the coastal waters near Anvers Island, Antarctica
Article Abstract:
The ecology of planktonic archaeal and bacterial assemblages in the waters of Anvers Island in Antarctica was investigated between 1995 and 1996. Ribosomal RNA hybridization revealed temporal as well as seasonal variations in the assemblages. Analysis by denaturing gradient gel electrophoresis of the Antarctic bacterioplankton further demonstrated this variability, with assemblages being stable in early spring and changing through the summer and with depth. In addition, variations in microbial assemblage composition were noted along chemical and physical gradients.

Rapid differentiation of bacterial species with multiple probes of different lengths in a single slot blot hybridization
Article Abstract:
A method using many oligonucleotide probes in a single slot blot hybridization is applied for the identification of bacterial species and the study of bacterial colonies. Probes of varying lengths are labeled with 32P and hybridized on a membrane containing genes coding for 16S rRNAs from several bacterial species. The bound probes are separated after hybridization and the genes are identified. This technique is economical and less time-consuming because it uses less reagents and exposure to radioactivity.

Application of rRNA-based probes for observing marine nanoplanktonic protists
Article Abstract:
The use of rRNA-based probes to observe marine protists was examined with a ciliate (an Uronema sp.), a flagellate (a Cafeteria sp.) and mixed assemblages of protists from enrichment cultures and natural water samples. The use of fluorescein- or biotin-labeled eukaryote-specific probes allowed detection of protists by epifluorescence microscopy. Moreover, the hybridization of natural assemblages of protists using biotinylated probes and fluorescent-labeled avidin was demonstrated.
