Sequence-resolved detection of pausing by single RNA polymerase molecules
Article Abstract:
An ultrastable optical-trapping assay is developed to follow the motion of individual molecules of ribonucleic acid polymerase (RNAP) transcribing templates engineered with repeated sequences carrying imbedded, sequence-specific pause sites of known regulatory function. The data obtained for the lifetimes and efficiencies of pauses support a model, where the transition to pausing branches off of the normal elongation pathway and is mediated by a common elemental state, which corresponds to the ubiquitous pause.

Predicting gene expression from sequence
Article Abstract:
The systematic genome-wide approach to learn complex combinatorial code underlying gene expression is explained using a probabilistic approach. It is found that for successful predicting one needs a complex set of rules that use logical reasoning with motif strength as well as significant constraints and relative position.
