Stable expression of pertussis toxin in Bordetella bronchiseptica under the control of a tightly regulated promoter
Article Abstract:
A minitransposon containing the tox operon, controled by a promoter responsive to an aromatic inducer, enhances the production of pertussis toxin (PT) by Bordetella bronchiseptica. Characterization of recombinant clones also reveals the tox operon under the control of the Psal promoter. The growth pattern of PT is independent of noninducing conditions and low inducer concentration. Thus, the strain performance is unaffected in bioreactors after uncoupling from gene expression. The biological activity of the recombinant PT is similar to the PT produced by B. pertussis.

Natural kirromycin resistance of elongation factor Tu from the kirrothricin producer Streptomyces cinnamoneus
Article Abstract:
Streptomyces cinnamoneus which produces the kirrothricin possess an elongation factor (EF) Tu resistant to kirromycin (Kr). The antibiotic kirromycin affects protein synthesis by inhibiting the release of EF-Tu to prevent elongation. The S. cinnamoneus single tuf gene was expressed in E. coli as a translational fusion to malE to evaluate the molecular determinants of kirromycin resistance. Thr was confirmed to be an essential determinant of Kr resistance in the naturally resistant protein of S. cinnamoneus.
