Structural basis of multidrug recognition by BmrR, a transcription activator of a multidrug transporter
Article Abstract:
The structural and stereochemical mechanisms in the multidrug recognition by the Bacillus subtilis transcriptor regulator BmrR was examined by crystallizing the BmrR C terminus complexed to the cationic drug tetraphenylphosphonium. The complex was found to result in a drug-induced unfolding and relocation of an alpha helix that led to the exposure of an internal drug-binding pocket. The tetraphenylphosphonium binding was found to be influenced by stacking and van der Waals contacts with multiple hydrophobic residues of the pocket and by the electrostatic interaction between the drug and a buried glutamate residue.

Three in one and one in three: it all depends on TBP
Article Abstract:
The TATA box-binding protein (TBP) is a general factor which plays a role in the formation of initiation complexes involving all three eukaryotic nuclear RNA polymerases. The role of TBP in TATA- and non-TATA-containing promoters is discussed. In general, TBP is characterized by its ability to bind to many other proteins including general factors, TBP-associated factors, activators and inhibitors. This versatility has been partially explained by the elucidation of TBP's three dimensional structure at a resolution of 2.6 angstroms.

The crystal structure of the IkappaBalpha/NF-kappaB complex reveals mechanisms of NF-kappaB inactivation
Article Abstract:
IkappaBs are a class of specific inhibitory proteins preventing nuclear translocation and DNA binding of the NF-kappaB transcription factor. Their structures manifest a centrally located ankyrin repeat domain and a signal receiving domain possessing amino acid residues. The basic mechanism involved in the NF-kappaB/IkappaBalpha regulation of transcription is the affinity of IkappaBalpha towards the p65 and c-Rel-containing homo- and heterodimers as well as the p50/p65 heterodimer.
