The Med proteins of yeast and their function through the RNA polymerase II carboxy-terminal domain
Article Abstract:
Mediator, resolved from Saccharomyces cerevisiae, is a multiprotein complex resolved based on the need for transcription activation in an entirely defined system. Three groups of mediator polypeptides were identified, products of five SRB genes, suppressors of carboxy-terminal domain (CRD)-truncation mutations and products of four genes, global repressors. Also there were six members of a new protein family, Med, deemed mainly responsible for transcription activation. Srbs 8, 9, 10, 11 and members of the SWI/SNF complex were absent from the purified mediator. Evidence exists for human homologs of several mediator proteins including Med7. That would indicate that like systems exist in higher cells.

Ubiquitylation of histone H2B controls RNA polymerase II transcription elongation independently of histone H3 methylation
Article Abstract:
The enzyme-dependent alterations in chromatin structure like ubiquitylation of histone H2Bthat leads to methylation of histone H3 are responsible for the transcription by RNA polymerase II. The authors report a divergence between ubiquitylation of H2B and methylation of Lys 4 on histone 3 in the chromatin of the budding yeast.

The human PAF complex coordinates transcription with events downstream of RNA synthesis
Article Abstract:
The isolation and functional characterization of the human PAF (hpaf) complex is reported. hPAF coordinates events in RNA quality control, in addition to coordinating events during transcription.
