The axonal chemoattractant netrin-1 is also a chemorepellent for trochlear motor axons
Article Abstract:
Examination of the dorsal migration of trochlear motor axons using explant culture of the entire hindbrain-midbrain junction from rat embryos revealed that floor plate cells repel trochlear motor axons at a distance in vivo. Movement was guided by netrin-1, a long-range chemoattractant that has been established to guide axons ventrally from the floor plate. Netrin-1 was observed to share functions with the UNC-6 protein found in C. elegans, which guides dorsal migrations. These results suggest that netrin-1 is a bifunctional long-range guidance cue that attracts ventrally directed axons and repels dorsally directed axons.

A ligand-gated association between cytoplasmic domains of UNC5 and DCC family receptors converts Netrin-induced growth cone attraction to repulsion
Article Abstract:
A study was conducted to determine the effect of the interaction of two families of receptors, namely, the Netrin receptors of the Deleted in Colorectal Cancer (DCC) and the UNC5 family receptors in axon guidance. Experiments with Xenopus spinal cord neurons revealed that the introduction of UNC5 proteins in cells transform the spinal axon-attraction of DCC netrin receptors to axon-repulsion. This is effected by the interaction of netrin-1 to UNC5.

Research reveals that besides the involvement of the chemoattractant Netrin-1 in the development of axons, the morphogen Sonic hedgehog mimics chemoattractant activity, suggesting that a morphogen can also serve as an axonal chemoattractant.
