Article Abstract:
The mechanism behind the interference of the growth factor-mediated induction of the Ras pathway on signaling through the second messenger cyclic AMP is studied. High stoichiometry resulted in the formation of the pp90rsk-CREB binding protein complex, which, when added with growth factor, persisted for 6-8 hours. Results indicate that cross-coupling between two signaling pathways can occur at the level of the signal-dependent coactivator.

Solution structure of the KIX domain of CBP bound to the transactivation domain of CREB: a model for activator:coactivator interactions
Article Abstract:
The activator:coactivator interaction between CREB and CREB-binding protein (CBP) promotes gene transcription at target segments of the DNA. This interaction is mediated at the KIX domain of CBP. The solution structure of this complex analyzed using nuclear magnetic resonance spectroscopy reveals that the CREB phosphorylated kinase inducible domain undergoes a transition from coil to helix during its interaction with KIX.

A pathogenic model of rheumatoid arthritis involves an E3 ubiquitin ligase, Synoviolin/Hrd1, as a causative factor for arthropathy. The enzyme initiates synovial cell outgrowth via antiapoptotic effects. Research indicates that synoviolin/Hrd1 can be a therapeutic target for rheumatoid arthritis.
