Two-dimensional crystallography of TFIIB- and IIE-RNA polymerase II complexes: implications for start site selection and initiation complex formation
Article Abstract:
The two-dimensional crystallographic structures of the transcription factors IIB (TFIIB) and TFIIE bound to RNA polymerase II indicate their importance in initiation-complex formation and start site selection. TFIIB brings the TATA DNA in contact with polymerase at a certain distance from the enzyme's active center. This distance defines the distance from the TATA box to the transcription start site. TFIIE participates in a key conformational switch occurring at the active center upon polymerase-DNA interaction.

Article Abstract:
A two-step mechanism of nucleotide selection is demonstrated, with initial binding to an entry (E) site beneath the active center in an inverted orientation, followed by rotation into the nucleotide addition (A) site for pairing with the template DNA. Findings indicate that the multisubunit RNA polymerases, typified by polymerase II, differ fundamentally from single subunit RNA and DNA polymerases.

Structural basis of transcription: Role of the trigger loop in substrate specificity and catalysis
Article Abstract:
The role of trigger loop in substrate specificity and catalysis as revealed by structures of transcribing complexes is reported. The trigger loop is found to swing beneath a correct nucleoside triphosphate (NTP) in the nucleotide addition site, closing off the active center and forming an extensive network of interactions with the NTP base, sugar, phosphates, and additional poll II residues.
