Unusual ribulose-1,5-biphosphate carboxylase/oxygenase genes from a marine manganese-oxidizing bacterium
Article Abstract:
Genes coding for both large (cbbL) and small (cbbS) subunits of a ribulose-1,5-biphosphate carboxylase/oxygenase (RubisCO) enzyme were identified, isolated and sequenced in the Gram-negative bacterium strain SI85-9A1. Introduction of these genes into an Escherichia coli host strain caused ribulose-1,5-biphosphate-dependent CO2 fixation under regulation of a lac promoter, suggesting that the protein is functonal in E. coli. Phylogenetic analysis of the RubisCO genes of strain SI85-9A1 indicated that they are divergent, but are more related to those from non-chlorophyte algal chloroplasts than are those from other bacteria.

The cytochrome c maturation operon is involved in manganese oxidation in Pseudomonas putida GB-1
Article Abstract:
Transposon mutagenesis was used to identify genes relevant to the manganese-oxidizing process in Pseudomonas putida GB-1. Several classes of mutants with differences in Mn oxidation were seen. Analysis of the non-oxidizing mutants suggested that a multicopper oxidase-related enzyme is involved in Mn oxidation. Two mutants defective in MN-oxidizing activity secreted porphyrins into the medium and appeared to be derepressed for pyoverdine production. Results indicated that a functional ccm operon is required for Mn oxidation in P. putida GB-1.

c-Type cytochromes and manganese oxidation in Pseudomonas putida MnB1
Article Abstract:
Mutants of Pseudomonas putida MnB1 that are unable to oxidize manganese were constructed and characterized using transposon mutagenesis. The mutants were divided into three groups which are mutants defective in the biogenesis of c-type chromosomes, mutants defective in genes that encode key enzymes of the tricarboxylic acid cycle and mutants defective in the biosynthesis of tryptophan. The first two mutants were cytochrome c oxidase negative and did not contain c-type cytochromes.
