ord1, an oxidoreductase gene responsible for conversion of omicron-methylsterigmatocystin to aflatoxin in Aspergillus flavus
Article Abstract:
The Aspergillus flavus (Af) ord1 oxidoreductase gene encoding a cytochrome (CYT) P450 monooxygenase that mediates the conversion of OMST or omicron-methylsterigmatocystin to aflatoxin was analyzed in yeasts cells. Molecular characterization of Af ord1 that were functionally expressed in Saccharomyces cerevisiae indicated the role of a gene encoding a 60.2-kDa CYT P-450 monooxygenase in mediating the conversion of OMST. Furthermore, the expression of ord1 was regulated by aflR which functions as a positive regulatory gene that encodes a zinc cluster DNA binding protein.

Identification of aflatoxin biosynthesis genes by genetic complementation in an Aspergillus flavus mutant lacking the aflatoxin gene cluster
Article Abstract:
Functional expression of aflatoxin biosynthesis genes in Aspergillus flavus 649 WAF2 transformants failed to exhibit aflatoxin production. Since strain 649 lacks a 90-kb genomic region, it was suggested that the gene cluster in the 90-kb region is involved in aflatoxin expression. Cloning and sequencing of the genes in the 90 kb of A. flavus genomic DNA detected avf1 and the oxidoreductase gene, ord1, indicating these these two genes are involved in aflatoxin biosynthesis.
