An engineered epigenetic transgene switch in mammalian cells
Article Abstract:
The two repressors that control of each other's expression are merged, then a mammalian epigenetic circuitry is formed that is capable to switch between the two stable transgene expression states once the two alternate drugs are temporarily administered. The epigenetic transgene control through the movable switches is an essential part in the engineering of artificial gene networks in mammalian cells.

Sperm derived from embryonic system (ES) cells can give rise to viable offspring mice that are abnormal and have imprinting defects. The high rate of abnormal development in these manipulated gametes is likely to be epigenetic is nature that involve DNA modification or chromatin modifications that in turn regulate gene expression essential for normal development.

The design and characterization of gas-inducible transgene control system functional in different mammalian cells, mice and prototype biopharmaceutical manufacturing are described. The acetaldehyde-inducible regulation (AIR) technology serves as a tool for therapeutic transgene dosing as well as biopharmaceutical manufacturing.
