High-throughput mapping of the chromatin structure of human promoters
Article Abstract:
A high-resolution microarray approach combined with an analysis algorithm to examine nucleosome positioning in 3,692 promoters within seven human cell lines is described. The combination of nucleosome data with chromatin immunoprecipitation-chip analyses reveals that the melanocyte master regulator microphthalmia-associated transcription factor (MITF) predominantly binds nucleosome-free regions, supporting the model that nucleosomes limit sequence accessibility.

Article Abstract:
Chromogenic stains, long used in immunodiagnostic assay, could probably be supplanted by fluorescence-based readouts as emphasis shifts away from diagnosis to prediction by means of quantifiable results. Fluorescence is a better mechanism to capture the advantages of multiplexing as pathologists move from immunohistochemistry into the next generation.
