Stable expression of shRNAs in human CD34(super +) progenitor cells can avoid induction of interferon responses to siRNAs in vitro
Article Abstract:
The immunostimulatory effects of lipid-delivered small interfering RNAs (siRNAs) versus Pol III promoter-expressed small hairpin RNAs (shRNAs) in primary CD34(super +) progenitor-delivered hematopoietic cells are investigated. The results have shown that in this system, lipid-delivered siRNAs are potent inducers of interferon (IFN) alpha (IFNalpha) and type I IFN gene expression, whereas the same sequences when expressed endogenously are nonimmunostimulatory.

An innovative screen that rapidly sorts thousands of environmental DNA clones to identify those with a desired metabolic activity is presented with the help of a designed high-throughput screen for catabolic pathways, substrate-induced gene expression screening' or 'SIGEX'. The SIGEX template allows infinite variation in the functions to be discovered, the regulator molecules to be employed and the reporter proteins to be detected.
