Synthetic dsRNA Dicer substrates enhance RNAi potency and efficacy
Article Abstract:
Synthetic RNA duplexes 25-30 nucleotides in length are found to be upto 100 fold more potent than corresponding conventional 21-mer small interfering RNAs (siRNAs). The enhanced potency of the longer duplexes is attributed to the fact that they are substrates of the Dicer endonuclease, directly linking the production of siRNAs to incorporation in the DNA-induced silencing complex.

A considerable improvement in siRNA delivery is achieved by covalently attaching cholesterol to one of the siRNA strands that allows direct intravenous low-pressure injection of the material into mice. RNA interference is a powerful cellular mechanism for targeted down regulation of gene expression.

Article Abstract:
Antisense agents used for sequence-specific knockdown of mRNA are antisense oligonucleotides (ODNs), ribozymes, DNAzymes and RNA interference. RNAi is unlikely to displace the use of antisense, ribozymes, DNAzymes and related approaches for many applications.
