Detection of viral deoxyribonucleic acid in the amniotic fluid of low-risk pregnancies by polymerase chain reaction
Article Abstract:
The DNA of various viruses may not be present in the amniotic fluid of pregnant women with low risks of viral infection. Researchers analyzed amniotic fluid samples from 277 pregnant women who were not at risk for viral infection but who underwent amniocentesis for older age or abnormal levels of alpha-fetoprotein. The polymerase chain reaction technique detected DNA in 87% of the samples but did not detect viral DNA in any of these samples. Detection of DNA from viruses such as adenovirus, cytomegalovirus, parvovirus, and enterovirus in amniotic fluid has been associated with a high risk of fetal infection that may in turn cause developmental abnormalities.

Specific detection of 16 micro-organisms in amniotic fluid by polymerase chain reaction and its correlation with preterm delivery occurrence

Polymerase chain reaction (PCR) gene amplification testing may be more sensitive than bacterial culturing in detecting microorganisms in the amniotic fluid of women in preterm labor with intact fetal membranes. Bacteria in the amniotic cavity may be involved in spontaneous preterm labor. Researchers compared PCR and culturing on amniotic fluid samples from 51 women in preterm labor and 23 pregnant women not in labor. PCR identified microorganisms in 46% of samples from women in labor, while culturing detected bacteria in only 12% of samples. No bacteria were found in samples from women not in labor.

Prenatal diagnosis of fetal varicella-zoster virus infection with polymerase chain reaction of amniotic fluid in 107 cases
Article Abstract:
A test used to identify genetic sequences called the polymerase chain reaction appears better able to detect possible fetal varicella zoster infections than amniotic fluid cell cultures or fetal blood tests. One hundred seven amniotic fluid samples from fetuses whose mothers had varicella zoster infections were analyzed using both cell culturing and polymerase chain reaction tests. Blood samples were also taken and tested. Positive test results were obtained in 8.4% of the polymerase chain reaction tests, 2.8% of the cell culture tests, and none of the blood tests.
