Sequence-specific potent induction of IFN-alpha by short interfering RNA in the plasmacytoid dendritic cells through TLR7
Article Abstract:
A demonstration that in plasmacytoid dendritic cells (PDC), an immune cell subset specialized in the detection of viral nucleic acids and production of type I interferon (IFN), some siRNA sequences, independent to their GU content, are potent stimuli of IFN-alpha production. Injection into mice of immunostimulatory siRNAs, when complexes with cationic liposomes, induced systemic immune responses, in the same range as the TLR9 ligand cpG, including IFN-alpha in serum and activation of T cells and dendritic cells in spleen.

An immune cell subset specialized in the detection of viral nucleic acids and production of type I interferon (IFN), some short interfering RNA (siRNA) sequences, are potent stimuli of IFN-alpha production in plasmacytoid dendritic cells (PDC). The sequence-specific Toll-like receptor 7 (TLR7)-dependent immune recognition in PDC should be considered as an additional biological activity of siRNA, which should be termed as immunostimulatory RNA (isRNA).

Therapeutic silencing of an endogenous gene by systemic administration of modified siRNAs
Article Abstract:
A demonstration that chemically modified short interfering RNAs (siRNAs) could silence an endogenous gene encoding apolipoprotein B (apoB) after intravenous injection in mice is presented. Administration of chemically modified siRNAs resulted in silencing of the apoB messenger RNA in liver and jejunum, decreased plasma levels of apoB protein, and reduced total cholesterol.
