Optimised sample preparation of synovial fluid for detection of Chlamydia trachomatis DNA by polymerase chain reaction
Article Abstract:
Optimal preparation of synovial fluid samples improves the diagnostic accuracy of polymerase chain reaction (PCR) DNA testing for Chlamydia trachomatis in reactive arthritis. Reactive arthritis is an inflammation of the synovial membranes of joints following an infection, often with the sexually transmitted bacterium Chlamydia trachomatis. Researchers processed synovial fluid samples containing the chlamydia organism in eight different ways to determine which method best improved PCR-based diagnostic testing. DNA isolation and extraction with cetyltrimethylammonium bromide was most effective in improving the diagnostic sensitivity of the test.

Direct detection of HIV-1 RNA in epidermal Langerhans cells of HIV-infected patients
Article Abstract:
HIV-1 RNA appears to be actively transcribed in Langerhans cells (LC). LCs are white blood cells derived from bone marrow cells that play a role in stimulating T-cells and initiating immune responses. Previous studies have suggested that HIV directly infects LCs. Skin slices from nine HIV-positive patients were compared with skin slices from 11 HIV-negative individuals. HIV-1 RNA was detected in the LCs of six of the HIV patients. HIV-1 RNA was not detected in any of the LCs from the HIV-negative individuals. These findings indicate not only that LCs are a site of HIV replication but also suggest that infected LCs transmit HIV to T cells.
