Application to environmental samples
Article Abstract:
Radioligand assay of environmental samples including fly ash, caustic ash, wash water and fish homogenates to mouse hepatic Ah receptor showed higher 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalent concentration values than those obtained by gas chromatography (GC) and mass spectrometry (MS). However, further analysis of the samples by GC and MS identified the additional Ah receptor-active material as coplanar polychlorinated biphenyl and polychlorinated aromatic hydrocarbons, indicating that Ah receptor assay is an effective method for determining the toxicity of dioxin-like compounds.

Competition assay of dioxin-like compounds and related analytes using fixed aliquots of radiolabeled 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and C57BL/6 mouse hepatic Ah receptor showed additive binding of various types of chlorinated aromatic compounds to the receptor. It also revealed a detection limit of 100 pg of TCDD equivalent when the radiolabel used was 3H TCDD. These results indicate that competition assay is more effective tool for prescreening environmental samples than conventional environmental analytical methods.

Article Abstract:
Louis P. Brzuzy and Ronald A. Hites' assumptions on the global deposition rates of polychlorinated dibenzo-p-dioxins dibenzofurans (PCDD/F) were erroneous. The global abundance of PCDD/F is due to the prevalence of brush and forest fires. Wood combustion produces significant amounts of PCDD/F together with volcanic eruptions and biological metabolism. Furthermore, natural biomass emission of PCDD/F is equivalent to five billion kilograms annually.
