Avoiding the road less traveled: how the topology of enzyme-substrate complexes can dictate product selection
Article Abstract:
Study reveals that two enzymes namely 1-Aminocyclopropane-1-carboxylate (ACC) synthase and 7,8-diaminopelargonic acid synthase are pyridoxal 5'-phophate-dependent enzymes that utilize S-adenosyl-L-methionine as a substrate but yield different products. The active-site topology of the enzyme-intermediate complexes helps in deciding this pathway bifurcation.

Complicated protein structures at high resolution is feasible to examine due to the recent advances in molecular cloning, protein expression, and X-ray crystallography. A tunnel for the migration of carbon monoxide is recently identified in the structure of carbon monoxide dehydrogenase/ acetyl-CoA synthase.

Biochemical and biophysical properties of the co-sensing transcriptional activator CooA

The molecular biological and spectroscopic studies, conducted to analyze the mechanisms of CO sensing by CooA and CO-dependent activation of CooA, are discussed. CooA is a CO-sensing transcriptional activator found in the photosynthetic bacterium Rhodospirillum rubrum.
