Neil Risch and Kathleen Merikangas conducted a series of studies on human disease-risk gene screening based on transmission-equilibrium test and sib-pair linkage analysis. Improvements, comments and suggestions to those studies are presented.

The future of genetic studies of complex human diseases
Article Abstract:
Association studies have greater potential than linkage studies in determining the genetic factors in complex diseases. Using association studies will require large-scale testing to identify the genes causing the disease.

The apparent chromosome locus identified in a study of Parkinson's disease (PD) may not be responsible for the majority of inherited idiopathic PD cases. Other studies indicate genetic heterogeneity in PD.
