A technical KO of amyloid-beta peptide
Article Abstract:
It has been established that the presenilin-1 (PS1) gene controls the unusual intramembranous proteolysis of the amyloid precursor protein (APP). It is therefore possible that PS1 could act as an unexpected target for drugs against Alzheimer's disease, an aggressive form of which is caused by mutations of the PS1 gene. Inhibition of presenilin synthesis late during ageing, when Alzheimer's disease normally begins, would avoid elimination of the vital function of presenilin proteins during development.

Secretion of beta-amyloid precursor protein cleaved at the amino terminus of the beta-amyloid peptide
Article Abstract:
Amyloid precursor protein (APP) gives rise to the beta-amyloid protein (A-beta) present in the senile plaques associated with Alzheimer's disease. Analysis of human brain cultures and cerebrospinal fluid shows that much of APP consists of a newly identified form separated at the alpha terminus of A-beta. Hence a secretory pathway may be implicated in producing A-beta, a finding that could lead to a better understanding of familial Alzheimer's.
