Limbs move beyond the Radical fringe
Article Abstract:
The mouse Rfng gene is not needed for limb development, even though it is expressed in the developing limb bud. A number of developmental defects are caused by the insertion of a selection cassette into the Rfng locus that has an impact on the expression of a neighbouring gene or genes. It is possible that the PGKneo cassette affects the expression of a neighbouring gene or genes, possibly paralogous between fringe family members. Other research in this area does not support the view that the presence of a neo-selection cassette inserted into the Rfng locus inevitably prompts embryonic lethality.

Article Abstract:
A vital genomic imprinting signal can be carried by differentially methylate regions in a specific imprinted gene, Igf2r, the maternally expressed gene that encodes the type-2 receptor for insulin-like growth factor. The imprinted gene loses its imprinting when the region is deleted from it, and the gene is expressed irrespective of parental origin. Methylation differences can be identified in two regions of the Igf2r gene. Region 1 is in the promoter, while region 2 is downstream in the second intron.
