Optical detection of a quantal presynaptic membrane turnover
Article Abstract:
Researchers have used the fluorescent dye FM 1-43 as a tracer for presynaptic endocytosis, thus making it possible to characterize both evoked and spontaneous components of presynaptic function at the level of single quanta. It was found that dye uptake takes place in quantal steps during evoked and spontaneous presynaptic function. There was seen to be temporal proximity of single-quantum dye uptake events to single-action potential stimuli. This indicates that unitary exocytic and endocytic events are closely connected in some way.

Clathrin self-assembly is mediated by a tandemly repeated superhelix
Article Abstract:
The cytoplasmic protein clathrin, polymerizes into a polyhedral lattices to form protein-coated membrane vesicles. The clathrin triskelion is a trimer of heavy-chain subunits each binding in the hub domain. The atomic structure of hub residues, 1,210-1,516, involved in mediating spontaneous clathrin heavy-chain polymerization and light-chain association, is reported. The hub fragment folds and a 145-residue motif is repeated seven times throughout the filamentous leg.

Beta-arrestin acts as a clathrin adaptor in endocytosis of the beta(sub-2)-adrenergic receptor
Article Abstract:
Beta-arrestin and arrestin-3 not only promote the internalization of beta(sub-2)-adrenergic receptots (B2AR) but binds directly and stoichiometrically to clathrin as well. Beta-arrestin/arrestin chimeras which have defects in B2AR or clathrin binding exhibit reduced ability to promote B2AR endocytosis. Beta-arrestin serves as an adaptor in the endocytosis pathway.
