Rhodopsin-family receptors associate with small G proteins to activate phospholipase D
Article Abstract:
The activation of phospholipase D that is induced by many, but not all, Ca2+-mobilizing G-protein-coupled receptors is sensitive to inhibitors of ARF and RhoA. These receptors contain the amino-acid sequence AsnProXXTyr in their seventh transmembrane domain, while receptors able to activate phospholipase D without involving ARF contain the sequence AspProXXTyr. One functional result of the association between certain rhodopsin-family receptors and the small G proteins ARF and RhoA seems to be an increased coupling of receptors to phospholipase D activation.

Phospholipase C isozymes selectively couple to specific neurotransmitter receptors
Article Abstract:
Researchers investigated the functions in the brain of two of the four isoenzymes of the enzyme phospholipase C (PLC). They studied the functions of PLC-beta1 and PLC-beta4 by generating null mutants in mice. The PLC-beta1 mice developed epilepsy and the PLC-beta4 mice demonstrated ataxia. The research revealed that PLC-beta1 affects signal transduction in the hippocampus and cerebral cortex, and that PLC-beta4 affects the metabotropic glutamate receptor in the cerebellum. These results indicate that neurological disorders have a pharmacological basis.

Involvement of Ral GTPase in v-Src-induced phospholipase D activation
Article Abstract:
The activation of phospholipase D (PLD) by v-Src is discussed in relation to the role of Ral GTPase. It had been earlier found out that PLD activity increases in response to tyrosine kinase activity in v-Src resulting in the production of such substances as diaglycerol and phosphatidic acid. Ral GTPase acted in a manner analogous to the activation by Ras of Ral-GDS.
