Uncoupling of S phase and mitosis induced by anticancer agents in cells lacking p21
Article Abstract:
Cells of the human colorectal cancer cell line HCT116 lacking the cyclin-dependent kinase inhibitor, p21, lead to defective cell cycles on treatment with DNA damaging anticancer agents. The cells arrest in the G2 phase of the cell cycle, then undergo rapid S phases without intervening mitosis and finally die. The cells are deformed and their nucleus is polyploid. This indicates that the block on DNA damaged cells entering mitosis is active in the cells but the block on S phases without an intervening M phase is inactive.

14-3-3delta is required to prevent mitotic catastrophe after DNA damage
Article Abstract:
An approach to the generation of human somatic-cell knockouts has been used to generate human colorectal cancer cells where both 14-3-3delta alleles are inactivated. The cells arrest initially after DNA damage, and 14-3-3delta -1- cells could not maintain cell-cycle arrest. The death of the 14-3-3delta -1- cells was linked to the failure of the 14-3-3delta-deficient cells in sequestering the proteins that initiate mitosis.
