Cloning and functional analysis of a phosphopantetheinyl transferase superfamily gene associated with jadomycin biosynthesis in Streptomyces venezuelae ISP5230
Article Abstract:
A phosphopantetheinyl transferase superfamily gene associated with jadomycin biosynthesis has been studied in Streptomyces venezuelae ISP5230. It has been shown that jadM, one of three complete ORRs found in the nucleotide sequence of its DNA, encodes a holo-ACP synthase needed mainly for jadomycin B. biosynthesis.

Accumulation of the angucycline antibiotic rabelomycin after disruption of an oxygenase gene in the jadomycin B biosynthetic gene cluster of Streptomyces venezuelae
Article Abstract:
The region of the Streptomyces venezuelae ISP5230 chromosome immediately downstream of the polyketide synthase gene cluster is investigated by cloning and sequence analyzing a fragment of the strain's DNA insert. Results indicate that the region encodes an oxygenase involved in opening ring B of an angucycline likely to be a biosynthetic intermediate. Jadomycins are formed when isoleucin molecules are injected into the site.

A role for pabAB, a p-aminobenzoate synthase gene of Streptomyces venezuelae ISP5230, in chloramphenicol biosynthesis
Article Abstract:
The p-aminobenzoic acid (PABA) synthase gene, pabAB, of Streptomyces venezuelae ISP5230 contributes to the synthesis of chloramphenicol (Cm). Disruption of the gene reduces Cm synthesis and resistance to sulfanilamide. The amino acid sequence of the cloned S. venezuelae DNA fragment indicates that the N- and C-terminal regions are similar to the PabA and PabB genes encoding PABA synthase enzymes in several bacteria. The sequence of the gene product is similar to the fused pabAB gene responsible for secondary metabolism in S. griseus.
