Enhancement of memory-related long-term facilitation by ApAF, a novel transcription factor that acts downstream from both CREB1 and CREB2

Research demonstrates that long-term memory storage related synaptic changes are regulated at the transcriptional level downstream from CREB1 in Aplysia. Results show that transcription activator ApAF facilitates long-term memory enhancement aided by serotonin and CREB1activation or ApCREB2 depression.

Aplysia CREB2 represses long term facilitation: relief of repression converts transient facilitation into long-term functional and structural change
Article Abstract:
Administration of anti-ApCREB2 antibodies into Aplysia sensory neurons causes a single pulse of serotonin which induces short-term facilitation to evoke long-term facilitation lasting more than one day. This facilitation requires transcription and translation, induces the growth of new synaptic connections and occludes further facilitation by five pulses of serotonin. The behavioral sensitization in Aplysia involves CREB-like proteins and immediate early gene ApC/EBP.

A transient, neuron-wide form of CREB-mediated long-term facilitation can be stabilized at specific synapses by local protein synthesis
Article Abstract:
A form of CREB-mediated long-term facilitation at specific synapses is discussed. It can be stabilized by local protein synthesis. Five pulses of 5-HT selectively applied to the soma of the bifurcated Aplysia sensory neuron give rise to the facilitation. It requires CREB1-mediated transcription, but it does not persist and is not growth-associated.
