Cap methyltransferase (MT) selective binding and methylation of GpppG-RNA are stimulated by importin-alpha according to this research communication. A human cDNA library has ben screened for mRNA cap methyltransferase-binding proteins. Nuclear transporter importin-alpha was isolated. Direct association was confirmed by several means.

Article Abstract:
Identification of a novel cellular mRNA-specific Internal ribosomal entry site (IRES)-specific cellular trans-acting factors (ITAF) is reported. The activities of Theiler's murine encephalomyelitis virus (TMEV) and foot-and-mouth disease virus (FMDV) IRESs were similar in RRL and in transfected BHK-21 cells. Replacement of the IRES of the neurotropic TMEV, however, by the IRES of the epitheliotropic FMDV gave viable but completely attenuated viruses that do not replicate in mouse neurons. Cap independent translation initiation on picornavirus mRNAs is mediated by an IRES in the 5' untranslated region. The needs for trans-acting factors are different in related picornavius IRESs. They can account for cell type-specific differences in IRES actions.
