Cloning, analysis and one-step disruption of the ARG5,6 gene of Candida albicans
Article Abstract:
The ARG5,6 gene from Candida albicans was isolated by complementation of a mutagenized C. albicans arginine auxotrophic strain present in strain EL2. Sequence analysis demonstrated a putative 857 amino acids (95 kDa) which was 63% identical to the Saccharomyces cerevisiae ARG5,6 gene. The gene which is similar to that of S. cerevisiae is responsible for the generation of a polypeptides that encode acetylglutamate kinase and acetylglutamyl-phosphate reductase activities in the arginine biosynthesis. An arg5,6 null mutant was generated in a single transformation event using the URA3-blaster protocol.

Phenotype in Candida albicans of a disruption of the BGL2 gene encoding a 1,3-beta-glucosyltransferase
Article Abstract:
Phenotypic changes occur in a Candida albicans strain which lacks 1,3-Beta-glucosyltransferase (BgI2p) due to disruption of both copies of the BGL2 gene. These changes show that cell wall alterations are found in Bgl2p-deficient strain CACW-1 which include attenuation of virulence as compared to the parent strain, increased sensitivity to nikkomycin Z, aggregation of cells during stationary phase and slower growth rate at 37 C for CACW-1 when compared to an isogenic parent.

Research demonstrates that resistance to fluconazole is mediated by FLU1 gene of the family of multidrug efflux transporters and disruptions in FLU1 enhanced the susceptiblity to the drug in Candida albicans.
