Construction of a 3-chlorobiphenyl-utilizing recombinant from an intergeneric mating
Article Abstract:
A recombinant 3- chlorobiphenyl-using strain was constructed from Pseudomonas specie strain HF1 and from Acinobacter specie strain P6. This strain was designated CB15. Hybridization experiments showed it was derived mainly from the Pseudomonas strain because of its physiological properties. Its ability to utilize 3- chlorobiphenyl, biphenyl and 3-chlorobenzoate as its carbon sources for growth did not extend to 3,3'-dichlorobiphenyl. Rate of 3-chlorobiphenyl biodegradation by this recombinant strain was greatly enhanced by increased surface area of the growth substrates. Construction of this catabolic phenotypewas done using a continuous amalgamated culture apparatus which has the abilityof selecting recombinants occurring at very low frequencies.

Metabolic and energetic control of Pseudomonas mendocina growth during transitions from aerobic to oxygen-limited conditions in chemostat cultures
Article Abstract:
An attempt was made to quantitate the control exerted on catabolic and anabolic fluxes of Pseudomonas mendocina growth during gradual transitions from aerobic to oxygen-limited conditions in continuous cultures. The results showed that under these conditions, P. mendocina undergoes transitions through three physiological phases. At high partial pressures of oxygen, the catabolism was mainly oxidative, with oxygen controlling catabolic fluxes. At lower partial pressures, oxygen controls both catabolic and anabolic fluxes. At microaerophilic conditions, mixed-type fermentations predominate.

Enhanced mineralization of polychlorinated biphenyls in soil inoculated with chlorobenzoate-degrading bacteria
Article Abstract:
The optimization of polychlorinated biphenyl (PCB) mineralization by the conjunction of chlorobenzoate degraders and PCB soil cometabolizers is discussed. This involved inoculation with chlorobenzoate utilizers, a biphenyl utilizer, combinations of the two physiological types and chlorobiphenyl-mineralizing transconjugants. Results indicate that biphenyl mineralization limit the chlorobenzoate metabolism in soils.
