Cell cycle- and cell growth-regulated proteolysis of mammalian CDC6 is dependent on APC-CDH1
Article Abstract:
Cell growth- and cell cycle-regulated proteolysis of mammalian CDC6 has been found to be dependent on APC-CDH1. CDC6 is targeted for ubiquitin-mediated proteolysis by the anaphase-promoting complex (APC)/cyclosome in G(sub.1). It is also not found in nonproliferating cells. Human CDC6 activity is regulated by CDK-regulated subcellular localization and periodic transcription. Overexpression of a mutant or wild-type CDC6 is not adequate to induce multiple DNA replications in the same cell cycle.

Regulation of centrosome duplication through the cell cycle, with consideration of the mechanisms and of coordination with nuclear events, is discussed in this review article. Topics include the role of centrosomes in determining spindle polarity, Cdks in the control of centrosome duplication, and ubiquitin-mediated proteolysis and the centrosome cycle.

Destruction box and KEN box motifs in budding yeast Hswl1p have been found to be necessary for anaphase-promoting complex (APC)-mediated degradation and direct binding to Cdc20p and Cdh1p. Regions of Hsl1p critical for binding to the APC machinery and for APC-mediated degradation have been studied.
