DNA topology and a minimal set of basal factors for transcription by RNA polymerase II
Article Abstract:
The minimal requirements for basal transcription from the immunoglobulin heavy chain (IgH) promoter were defined to be TATA-binding protein, TFIIB, and RNA polymerase II. The RNA synthesis in this reconstitution reaction required that the DNA template be negatively supercoiled. The findings suggest that the free energy of supercoiling promotes the formation of an open complex for initiation of transcription by the minimal set of transcription factors.

Transcriptional fidelity and proofreading by RNA polymerase II
Article Abstract:
The proofreading ability of the 3' to 5' nuclease activity of human RNA polymerase II is enhanced in the presence of a protein which stimulates the activity of nucleases, SII. Misincorporated bases are removed during the rapid chain elongation. These bases are identified during the slow incorporation of the succeeding base to the mismatched terminus. The mechanism of discrimination between correct and incorrect nucleotides is discussed.

RNA helicase A mediates association of CBP with RNA polymerase II
Article Abstract:
CBP, a coactivator, is thought to stimulate signal-dependent gene expression, via a link with RNA polymerase II complexes. The complex formation between CBP and RNA polymerase II was shown to need RNA helicase A (RHA), related to the Drosphila male dosage compensation factor mle. RHA cooperated with CBP in target gene activation mediation through CREB, the CAMP responsive factor.
