Detection of methanotrophic bacteria in environmental samples with the PCR
Article Abstract:
Experimental studies help design PCR primers for four structural genes in the soluble methane monooxygenase gene clusters of various methanotrophs and for the gram-negative methylotroph-specific methanol dehydrogenase gene moxF. A combined hybridization PCR assay helps confirm the specificity of these primers. Experimental studies use these primers to detect methanotroph DNA in environmental samples. Peat samples have a large number of distinct methanotrophs. Sequence analysis reveals that the mmoC gene exhibits significant level of variability, and the mmoX gene is significantly conserved in methanotrophs.

Methanol improves methane uptake in starved methanotrophic microorganisms
Article Abstract:
Methanotrophs sustain high rates of atmospheric methane oxidation if they are supplied with methanol or formate. Without being supplied by methanol or formate, their atmospheric methane oxidation stops but is reactivated within hours in response to methanol or formate. In the case of indigenous forest soil methanotrophs, a dependence on methanol supply upon reduced methane access is noted, but only upon exposure to a methane-free atmosphere. They respond to each methanol addition by halting atmospheric methane oxidation and restoring this methane oxidation as the methanol is depleted.
