Article Abstract:
Significant breakthroughs in biophysics and macromolecular physiological research have improved information on the cellular mechanisms of muscle fiber motility. The sliding filament theory on the intercation of actin and myosin during muscle contraction was supplemented by atomic analysis of myosin structure or actomyosin complexes and in vitro motility analysis. Analysis of the myosin mechanochemical cycle in skeletal muscle fibers also provided vital information on myosin motility and crystal structure.

Simultaneous observation of individual ATPase and mechanical events by a single myosin molecule during interaction with actin
Article Abstract:
Research was conducted to describe a method for measuring individual ATPase and mechanical reactions of single myosin molecules during force generation. The technique, which permits mechanical and ligand-binding events in a single myosin molecule, characterizes the ATPase reaction associated with the mechanical reaction in the myosin molecular motor. The assay could also be extended to investigate other allosteric enzymes and examine high energy states or signals after ligand changes.

Nuclear movement regulated by Cdc42, MRCK, myosin, and actin flow establishes MTOC polarization in migrating cells
Article Abstract:
The direct imaging of wound-edge fibroblasts after triggering microtubule-organizing center (MTOC) reorientation with soluble factors showed that the nucleus moved away from the leading edge to reorient the MTOC, while the MTOC remained stationary. The nuclear repositioning is an initial polarizing event in migrating cells and separate regulatory pathways establish the positions of the nucleus and the MTOC.
