Cloning of a phenol oxidase gene from Acremonium murorum and its expression in Aspergillus awamori

Researchers have cloned a phenol oxidase from the fungus Acremonium murorum in Saccharomyces and Aspergillus that might be used in laundry cleaning solutions. It is active at high temperatures and alkaline pH and is capable of decolorizing plant chromophores.

Analysis of heterologous protein production in defined recombinant Aspergillus awamori strains
Article Abstract:
The levels of steady-state mRNA of many heterologous genes of different origin vary widely. The values are high for genes of fungal origin and low for those of nonfungal origin. Researchers compared mRNA levels and protein levels of the genes controlled by the Aspergillus awamori 1,4-beta-endoxylanase A expression signals and integrated in a single copy at the A. awamori pyrG locus. The protein levels compare well with those predicted from the mRNA levels except for human interleukin 6. The protein levels are probably low for interleukin 6 due to intracellular protein degradation.

Glucoamylase gene fusions alleviate limitations for protein production in Aspergillus awamori at the transcriptional and (post)translational levels
Article Abstract:
The fusion of the Aspergillus niger glucoamylase gene (glaA) to the human interleukin-6 gene (hil6) or a synthetic guar alpha-galactosidase (aglA) gene removes the transcriptional and (post-)translational block on formation of AGL and hIL -6 mRNA and protein. The increase in protein levels is due to an increase in the steady-state mRNA level. The fusion of glaA to the wild-type aglA produces truncated protein due to defective processing of the aglA in the nucleus.
