Researchers describe a PCR-based method that can distinguish human fecal contamination from cow fecal contamination. It can also be used to identify bacterial contamination in complex environments and does not rely on cultures.

A PCR assay to discriminate human and ruminant feces on the basis of host differences in Bacteroides-Prevotella genes encoding 16S rRNA

Research describes the development of a polymerase chain reaction based assay for the detection of Bacteroides-Prevotella in human and ruminat feces. The16S rDNA clones from natural waters, identified by the bacterial rDNA PCR markers distinguishing cow from human feces, provide sequences for the development of specific PCR primers.

Host distributions of uncultivated fecal bacteroidales bacteria reveal genetic markers for fecal source identification
Article Abstract:
Bacteriodales 16S rRNA gene sequences from the feces of eight hosts were analyzed. The purpose of this study was to examine host distribution patterns among fecal bacteria in the order Bacteroidales, with the goal of using endemic sequences as markers for fecal source identification in aquatic environments.
