In vivo transposon mutagenesis in Haemophilus influenzae
Article Abstract:
A study was conducted to characterize an in vivo insertion mutagenesis in Haemophilus influenzae. The Qiagen kit protocol was utilized to carry out plasmid DNA preparation while the insertion sites of the Tn10 minitransposon elements were obtained using the dideoxynucleotide chain termination technique. A set of plasmids were then used to integrate minitransposon elements into H. influenzae. Results indicated that mini-Tn10 transposons can be utilized for the in vivo mutagenesis of H. influenzae.

Construction of a large signature-tagged mini-tn5 transposon library and its application to mutagenesis of Sinorhizobium meliloti
Article Abstract:
A signature-tagged transposon mutagenesis method was used that allowed analysis of the survival and the competitiveness of many mutants in a single experiment. In order to generate defined, nonredundant sets of signature-tagged Sinorhizobium meliloti mutants for subsequent experiments, 12,000 mutants were constructed, and insertion sites for more than 5,000 mutants were determined.

In vivo random mutagenesis of Bacillus subtilis by use of TnYLB-1, a mariner-based transposon
Article Abstract:
The construction and characterization of a mariner-based transposon system designed to be used in Bacillus subtilis is reported. It is observed that the transposon delivery system may be useful for insertional mutagenesis in other gram-positive bacteria.
