The sequence of spacers between the consensus sequences modulates the strength of prokaryotic promoters
Article Abstract:
A library of synthetic promoters for Lactococcus lactis was studied to determine whether sequences of spacers between consensus sequences regulates the strength of the promoters. The study reveals that the promoters, the strongest of which can exceed 2,000 relative units, can also modulate gene expression in Escherichia coli. DNA sequencing revealed that the weaker promoters exhibited changes either in the consensus sequences or in the length of the spacer between the -35 and -10 sequences.

Cloning and expression of the Lactococcus factis purDEK genes, required for growth in milk
Article Abstract:
The purDEK operon from the Lactococcus lactis subsp. lactis strain CHCC285 was cloned and its nucleotide sequence was characterized. Three reading frames showed substantial similarity with other organisms. The expression of the purDEK operon was observed to be controlled at the transcriptional level, providing about 35 times more expression in the absence of purines in the growth medium. The purD promoter and regulatory region was found to be located between nucleotides 450 and 593.
