Know your neighbors: three phenotypes in null mutants of the myogenic bHLH gene MRF4
Article Abstract:
The effects of gene disruption on the expression of other genes were studied using the myogenic basic helix-loop-helix (bHLH) gene MRF4. Three mutations were induced in the MRF4 to eliminate its function. Results showed that disruption of the MRF4 also affected several genes that were located at or nearby the affected loci. Disruption of the MRF4 affected a cis-acting regulatory function located at the MRF4/Myf5 locus. Precautions, such as ensuring definitive knowledge of the genes located at a certain locus, should be taken to ascertain the results of gene disruption studies.

I-mf, a novel myogenic repressor, interacts with members of the MyoD family
Article Abstract:
The mechanism by which myogenic factors of the MyoD family in the dermomyotome are regulated during somitogenesis was investigated using a yeast two-hybrid screen. A novel myogenic repressor, I-mf, was isolated and shown to block transcriptional activation of MyoD family members, which results in the prevention of myogenesis. Immunoprecipitation assay showed that I-mf exerts its action by interfering with MyoD family members' DNA binding activity or by retaining MyoD family members in the cytoplasm by masking their nuclear localization signals.

Pax7 is required for the specification of myogenic satellite cells
Article Abstract:
Pax, a paired-box transcription factor, has been found to be necessary for the specification of myogenic satellite cells. It was isolated by representational difference analysis as a gene specifically expressed in cultured satellite cell-derived myoblasts. It was surprising that fluorescence-activated cell-sorting analysis showed that the size of the fraction of muscle-derived stem cells was not affected. Results show muscle-derived stem cells and satellite cells are distinct from one another as cell populations.
